Bone metastasis: mechanisms, therapies, and biomarkers

چکیده
Skeletal metastases are frequent complications of many cancers, causing bone (fractures, pain, disability) that negatively affect the patient’s quality life. Here, we first discuss burden skeletal in cancer metastasis. We then describe pathophysiology Bone metastasis is a multistage process: long before development clinically detectable metastases, circulating tumor cells settle and enter dormant state normal vascular endosteal niches present marrow, which provide immediate attachment shelter, only become active years later as they proliferate alter functions bone-resorbing (osteoclasts) bone-forming (osteoblasts) cells, promoting destruction. The molecular mechanisms involved mediating each these steps described, also explain how interact with myriad interconnected cell populations including rich network, immune adipocytes, nerves. metabolic programs could engage to specifically grow bone. progress future directions existing bone-targeted agents report emerging therapies have arisen from recent advances our understanding metastases. Molecular Mechanisms of Bone Metastasis.
Metastasis of breast and prostate cancer as well as multiple myeloma to the bones represents a significant medical problem. We herein discuss the molecular basis of the creation of pre-metastatic niches, the process of bone metastasis and the phenomenon of tumor dormancy in the bone marrow as well as its regulation. We describe the identification and validation of genes mediating bone metastasi...

Cancer metastasis to the bone develops commonly in patients with various malignancies, and is a major cause of morbidity and diminished quality of life in many affected patients. Emerging treatments for metastatic bone disease have arisen from advances in our understanding of the unique cellular and molecular mechanisms that contribute to the bone metastasis. The tendency of cancer cells to met...
متن کاملMechanisms of bone invasion and metastasis in human neuroblastoma.
Bone is the second most common site of metastasis in neuroblastoma. Over the last several years, our understanding of the mechanism of bone metastasis in neuroblastoma has significantly improved. Like breast cancer and myeloma, neuroblastoma cells activate osteoclasts to form osteolytic lesions. Activation occurs via the receptor activator of NFkappaB ligand (RANKL) or in the absence of RANKL v...
متن کاملBone metastasis in prostate cancer: molecular and cellular mechanisms (Review).
Prostate cancer is the most common cancer in men in Western countries, with a high incidence of bone metastasis. The bony metastasis is incurable and contributes significantly to disease-specific morbidity and mortality. The molecular and cellular mechanisms leading to the development of bone metastasis in prostate cancer remain unclear, but are currently under intensive investigation. In this ...
متن کاملMolecular mechanisms of bone metastasis and associated muscle weakness.
Bone is a preferred site for breast cancer metastasis and leads to pathologic bone loss due to increased osteoclast-induced bone resorption. The homing of tumor cells to the bone depends on the support of the bone microenvironment in which the tumor cells prime the premetastatic niche. The colonization and growth of tumor cells then depend on adaptations in the invading tumor cells to take adva...
